基于AFLP分子标记技术的黑花韭与多星韭遗传多样性与亲缘关系分析

摘要:
采用AFLP分子标记技术分析了分布在滇中、滇西、滇西北的3个黑花韭居群、3个二倍体多星韭居群和6个四倍体多星韭居群的遗传多样性与亲缘关系。结果表明,12个居群在DNA水平上已发生明显分化。居群间的基因分化系数(GST)为0.444。由GST计算的居群间基因流(Nm)为0.625,表明居群间部分基因流动受阻。12个居群间的遗传距离(D)在0.019~0.298之间,平均为0.185。UPGMA聚类结果表明居群聚类关系与地理分布相近似。四倍体多星韭与黑花韭的遗传距离较与二倍体多星韭的遗传距离要小。二倍体多星韭与黑花韭的遗传距离最大,它们之间的亲缘关系最远。

Abstract:
Genetic diversity and relationship of 12 populations of Allium tchongchanense and A.wallichii which distributed in the central region,western region and northwestern region of Yunnan Province were analyzed by AFLP. The results showed that there was obvious genetic differentiation among populations. Genetic differentiation coefficient GST=0.444. Nm=0.625 calculated by GST showed that gene flow was interrupted among populations. The genetic distance(D)of 12 populations was 0.019-0.298,average 0.185. UPGMA cluster results showed that genetic distance relationship was in accordance with geographic distribution. The genetic distance between A.wallichii tetraploid and A.tchongchanenseow was smaller than that between A.wallichii tetraploid and A.wallichii diploid. The genetic distance was the largest between A.wallichii diploid and A.tchongchanense.
