HPLC法测定牡荆中牡荆素的含量

摘要:
建立了牡荆中牡荆素含量的高效液相色谱测定方法,对牡荆资源进行评价,为牡荆的质量标准的建立提供参考。样品采用超声波提取法提取,色谱柱为菲罗门柱(5 μm,250 mm×4.6 mm),流动相为甲醇-水溶液(4:6),流速为1.0 mL·min-1,检测波长为340 nm,进样量为5 μL,在柱温30 ℃时对牡荆中牡荆素的含量进行检测。结果表明,待测组分与其他组分分离度良好,牡荆素在0.04520~0.4068 μg范围内呈良好的线性关系,r=0.9998; 平均回收率为100.4%,RSD为1.63%。该方法简便、准确、重现性好,适用于牡荆中牡荆素的含量测定及药材质量控制。

Abstract:
A method was established for determining contents of vitexin in Vitex negundo var.cannabifolia(Sieb.et Zucc.)by HPLC in order to evaluate this resource. Vitexin was obtained by ultrasonic extraction,separated on Filo door column(5 μm,250 mm×4.6 mm)with a mixture of methanol- water(volume ratio 4:6)as mobile phase at flow rate 1.0 mL·min-1. The UV detective wavelength was 340 nm,injection volume was 5 μL,and the column temperature was set at 30 ℃. Vitexin content showed a good linear relationship within 0.04520-0.4068 μg.(r=0.9998)and the average recovery was 100.4%,RSD was 1.63%. This method is simple,accurate and can be used for quality control of Vitex negundo var.cannabifolia.
