建立了高效液相色谱测定罗汉果根中两个结构新颖的降三萜糖苷,罗汉果酸苷乙Ⅱ(Siraitic acid ⅡB)和罗汉果酸苷甲Ⅱ(Siraitic acid ⅡA)含量的方法。色谱条件:色谱柱:ZORBAX SB-C18柱(4.6 mm×150 mm,5 μm); 柱温:25 ℃; 流动相:乙腈-水(梯度洗脱:0→4 min:35%乙腈; 4→16 min:35%→45%乙腈); 流速:0.5 mL/min; 检测波长:230 nm; 进样量:10 μL。结果表明,罗汉果酸苷乙Ⅱ在0.05~0.3 mg/mL,罗汉果酸苷甲Ⅱ在0.05~0.25 mg/mL之间线性关系良好。该方法稳定,精密度、重现性好,且样品处理简单,提取与检测时间短。 |

A HPLC method was developed for determining the contents of Siraitic acid ⅡB and Siraitic acid ⅡA,which were novel 29-norcucurbitane triterepene glycosides in the roots of Siraitia grosvenorii. The chromatographic analysis was carried out on a ZORBAX SB-C18 column(4.6 mm×150 mm,5 μm),and the mobile phase was acetonitrile(A)-water(gradient elution:0→4 min:0→35%A,4→16 min:35%→ 45%A). The flow rate was maintained at 0.5 mL/min. The detection wavelength was set at 230 nm,and the column temperature was controlled at 25 ℃. The sample injection volume was 10 μL. The results indicated that Siraitic acid ⅡB had a linear relationship at the range of 0.05-0.3 mg/mL,and Siraitic acid ⅡA had a linear relationship at the range of 0.05-0.25 mg/mL. This method was accurate,stable and reproducible,while the preparation of samples was simple and detected time was short. |
