以梨蒴珠藓无菌藓株为外植体诱导愈伤组织和配子体再生,接种于含不同激素组合的MS和Knop固体培养基上,分别进行愈伤组织和不定芽的分化,并探讨愈伤组织诱导和配子体再生的适宜培养条件。结果显示,愈伤组织诱导的最佳培养基是MS+0.5 mg/L BA+0.1 mg/L 2,4-D,愈伤组织诱导率为33.3%; 不定芽诱导的最佳培养基是MS+1.0 mg/L BA,增殖系数为10.0; 不含激素的基本培养基有利于配子体的再生。该研究旨在筛选出梨蒴珠藓愈伤组织的诱导体系,并为其在系统分类上提供参考依据。 |

This research was conducted to select a suitable culture system for the callus induction and gametophyte regeneration of Bartramia pomiformis. And axenic mosses were inoculated on MS and Knop solid media with different hormone combilations for callus and buds induction and differentiation. The result showed that MS medium containing 0.5 mg/L BA,0.1 mg/L 2,4-D was optimal for callus induction,with the induction rate of 33.3%. Optimal medium for inducing gametophyte is MS+1.0 mg/L BA,and the achieved multiplication coefficient was 10.0. And Knop with no hormone was as well as MS in generation of gametophyte. This investigation could provide some important references for callus induction and systematics of B.pomiformis. |
