采用HPLC法对不同产地的苦玄参药材主成分苦玄参苷ⅠA和ⅠB的含量进行分析,发现苦玄参苷ⅠB含量高于苦玄参苷ⅠA; 色谱条件:Waters C18(4.6 mm i.d.×250 mm,5 μm)色谱柱,流动相:乙睛:水=36︰64(体积比),流速为0.8 mL/min,检测波长为264 nm。苦玄参苷ⅠA、ⅠB分别在0.05~0.50 mg/mL和0.072~0.720 mg/mL与峰面积有良好的线性关系。苦玄参苷ⅠA的线性回归方程Y=13 658X-92.72(r=0.999 3),平均回收率为100.58%(n=5),相对标准偏差(RSD)为1.86%; 苦玄参苷ⅠB的线性回归方程Y=5 258.9X-41.01(r=0.999 4),平均回收率为101.15%(n=5),相对标准偏差(RSD)为1.31%。该研究为制定苦玄参药材质量检测标准提供了简便、快速、准确的方法。 |

The content of picfeltarraenins ⅠA and ⅠB in Picria fel-tarrae from different regions were analyzed by HPLC,the results showed that the content of picfeltarraenins ⅠB was higher than picfeltarraenins ⅠA. The chromatography conditions were as follows:Waters C18 column(4.6 mm i.d.×250 mm,5 μm)with acetonitrile-water(36︰64,volume ratio)as mobile phase. The flow rate was 0.8 mL/min,and the detection wavelength was set at 264 nm. The operating curves of ⅠA and ⅠB were found to be linear over the ranges of 0.05-0.50 mg/mL(r=0.999 3)and 0.072-0.720 mg/mL(r=0.999 4). The average recoveries were 100.58% and 101.15%. The method is simple,accurate and can be used as a standard for the Picria fel-tarrae medicine material quality detected. |
