红芽大戟愈伤组织诱导及分化研究

摘要:
以野生红芽大戟茎段、叶片和嫩芽为材料,经消毒灭菌后,接种于含不同激素组合的MS固体培养基上,分别进行愈伤组织诱导和分化,结果表明,在茎段、叶片和嫩芽3种外植体中,嫩芽的愈伤组织诱导率最高,生长最好,最佳诱导愈伤组织激素组合为MS+6-BA2.0 mg·L-1+NAA0.4 mg·L-1。该研究旨在筛选出适合红芽大戟诱导分化的培养体系,进一步解决生产中繁殖率低的问题,为红芽大戟组织培养快速繁殖、种质资源保存提供重要参考依据。

Abstract:
This research was conducted to select a suitable culture system for the induction and differentiation of Knoxia valerianoides,so as to improve the propagation rate during its production. After disinfection and sterilization,the stems,leaves and buds as the experimental materials were inoculated on MS solid media with different hormone combinations for callus induction and differentiation. The results showed that the buds had the highest inductivity and the best growth among the three explants(stem,1eaf and bud). The best hormone combination for callus induction was MS medium supplemented with 2.0 mg/L 6-BA and 0.4 mg/L NAA. This investigation could provide some important references for the rapid propagation of tissue culture and germplasm resource conservation of K.valerianoides.
