拟南芥干旱胁迫诱导型启动子RD29B驱动AtCKX1基因植物表达载体的构建

摘要:
从拟南芥基因组中分别克隆AtCKX1基因和RD29B基因5′ 侧翼1705 bp启动子区域序列,生物信息学表明,AtCKX1含有黄素腺嘌呤二核苷酸(FAD)和细胞分裂素的结合位点;RD29B启动子片段中存在ABA响应元件(ABA response element;ABRE)、Myb结合位点、TATA 盒、CAAT 盒等顺式作用元件。分别将AtCKX1和RD29B插入载体pCAMBIA1390,构建了由RD29B驱动的AtCKX1的植物双元表达载体p1390RD29BAtCKX1。

Abstract:
In this study,the cloning of AtCKX1 and 5′ flanking promoter sequence of drought inducible RD29B from Arabidopsis was reported. The 1705 bp promoter sequence was analyzed bioinformatically in the database of plant cis acting regulatory element (PlantCARE). The result showed that there were several important cis acting elements,including ABRE (ABA response element),Myb binding site,TATA box,CAAT box,in the 1705 bp promoter region. The binary vector harboring AtCKX1 driven by RD29B promoter was further constructed to be used in coming plant transgene.
